چکیده

Attempts to induce liver tumors with the car cinogenic polycyclic aromatic hydrocarbons have generally failed (9). However, certain of these hydrocarbons have a marked effect on hepatic carcinogenesis induced by other agents. Thus, Richardson et al. (32) observed that 3-methylcholanthrene1 (MC), administered at a low level to rats also receiving the strong hepatocarcinogen 3'-methyl-4-dimethylaminoazobenzene(3'-methyl-

Hypothetical Two-Step Initiation of Experimental Carcinogenesis by Polycyclic Aromatic Hydrocarbons and Aminoazo Dyes

A new hypothesis is discussed, which describes the initiation of the carcinogenesis through polycyclic aromatic hydrocarbons (PAHs) and aminoazo dyes (AZOs) as a two-step process: the oncogenic proteins of the ras or ras-like on-cogenes activated by mutation ("initiation A ") co-operate with the complexes in the plasma membrane formed during the "initiation B " stage from the parent compounds o...

gens in the neoplastic processes induced by these compounds. The fact that the aromatic aminoazo dyes are carcinogenic primarily in the liver and generally not at sites of local application (16) has suggested that the aminoazo dyes are likewise active through the formation of reactive metabolites. The structural similarity be tween the aminoazo dyes and the carcinogenic aromatic amines and amid...

Anticarcinogenic activities of organic isothiocyanates: chemistry and mechanisms.

Organic isothiocyanates block the production of tumors induced in rodents by diverse carcinogens (polycyclic aromatic hydrocarbons, azo dyes, ethionine, N-2-fluorenylacetamide, and nitrosamines). Protection is afforded by alpha-naphthyl-, beta-naphthyl-, phenyl-, benzyl-, phenethyl-, and other arylalkyl isothiocyanates against tumor development in liver, lung, mammary gland, forestomach, and es...

Analysis of lipophilic carcinogen-membrane interactions using a model human erythrocyte membrane system.

Human erythrocytes have been used as a model for evaluating the chemical carcinogen-plasma membrane interaction. The carcinogenic aromatic amines 2-acetylaminofluorene, dimethylaminoazobenzene, and 3'-methyldimethylaminoazobene stabilize erythrocytes against lysis in hypotonic solution. In general, the stabilization potential of these compounds reflects their oil:water partition coefficients an...

Epigenetic effectiveness of complete carcinogens: specific interactions of polycyclic aromatic hydrocarbons and aminoazo dyes with cholesterol and apolipoprotein A-I.

During a co-precipitation of cholesterol (Chol) and slight amounts of polycyclic aromatic hydrocarbons (PAHs) or aminoazo dyes (AZOs) in aqueous albumin solution, complex particles are formed; on their surfaces apolipoproteins with an amphipathic alpha-helix (e.g. apoA-I) are more or less firmly adsorbed. An efficacy index can be calculated from the strength of the hydrophobic interactions betw...
